Monkey Cluster of Differentiation 68 ELISA Kit
Principle of the Assay: CD6 ELISA kit applies the competitive enzyme immunoassay technique utilizing a polyclonal anti-CD6 antibody and an CD6-HRP conjugate. The assay sample and buffer are incubated together with CD6-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the CD6 concentration since CD6 from samples and CD6-HRP conjugate compete for the anti-CD6 antibody binding site. Since the number of sites is limited, as more sites are occupied by CD6 from the sample, fewer sites are left to bind CD6-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The CD6 concentration in each sample is interpolated from this standard curve.

This gene encodes a 110-kD transmembrane glycoprotein that is highly expressed by human monocytes and tissue macrophages. It is a member of the lysosomal/endosomal-associated membrane glycoprotein (LAMP) family. The protein primarily localizes to lysosomes and endosomes with a smaller fraction circulating to the cell surface. It is a type I integral membrane protein with a heavily glycosylated extracellular domain and binds to tissue- and organ-specific lectins or selectins. The protein is also a member of the scavenger receptor family. Scavenger receptors typically function to clear cellular debris, promote phagocytosis, and mediate the recruitment and activation of macrophages. Rapid recirculation of CD68 from endosomes and lysosomes to the plasma membrane may allow macrophages to crawl over selectin-bearing substrates or other cells.
