Optimized DNA sequence encoding Human Tissue Plasminogen Activator mature chain was expressed in Chinese Hamster Ovary cells.
Molecular Weight
Native human Tissue plasminogen Activator,generated by the proteolytic removal of the signal peptide and propeptide, has a calculated molecular mass of approcimately 55 kDa. Recombinant tPA is a disulfide-linked monomer protein consisting 527 amino acid residue subunits, and migrates due to glycosylation as an approcimately 60 kDa protein under non-reducing conditions in SDS-PAGE.
Endotoxin
Endotoxin content was assayed using a LAL gel clot method.
Endotoxin level was found to be less than 0.1 ng/ug (1EU/ug).
Reconstitution
A quick spin of the vial followed by reconstitution in distilled water to a concentration not less than 0.1 mg/mL. This solution can then be diluted into other buffers.
Preparation and Storage
The lyophilized protein is stable for at least 2 years from date of receipt at -20°C.
Upon reconstitution, this cytokine can be stored in working aliquots at 2-8°C for one month, or at -20°C for six months, with a carrier protein without detectable loss of activity.
Avoid repeated freeze/thaw cycles.

CD40 protein is expressed on all B-lymphocytes during various stages of development, activated T- cells and monocytes, follicular dendritic cells, thymic epithelial cells, and various carcinoma cell lines. It is expressed on most mature B-cell malignancies and on some early B-cell acute lymphocytic leukemias. Human CD40 has been found to be the receptor for a protein involved in an X- linked immunodeficiency syndrome. The CD40 ligand is identical with a T-cell restricted cell surface activation antigen designated 5c8. The mouse ligand of CD40 is gp39.Based on its homology with other members of the TNF ligand protein superfamily the protein is referred to also as TNFSF5.

NCBI Description

The protein encoded by this gene is expressed on the surface of T cells. It regulates B cell function by engaging CD40 on the B cell surface. A defect in this gene results in an inability to undergo immunoglobulin class switch and is associated with hyper-IgM syndrome. [provided by RefSeq, Jul 2008]

CD40LG: Mediates B-cell proliferation in the absence of co- stimulus as well as IgE production in the presence of IL-4. Involved in immunoglobulin class switching. Defects in CD40LG are the cause of X-linked immunodeficiency with hyper-IgM type 1 (HIGM1); also known as X-linked hyper IgM syndrome (XHIM). HIGM1 is an immunoglobulin isotype switch defect characterized by elevated concentrations of serum IgM and decreased amounts of all other isotypes. Affected males present at an early age (usually within the first year of life) recurrent bacterial and opportunistic infections, including Pneumocystis carinii pneumonia and intractable diarrhea due to cryptosporidium infection. Despite substitution treatment with intravenous immunoglobulin, the overall prognosis is rather poor, with a death rate of about 10% before adolescence. Belongs to the tumor necrosis factor family.
