Intended Uses: This immunoassay kit allows for the in vitro quantitative determination of human programmed cell death 1 ligand 1,PDCD1 ligand 1 concentrations in serum, Plasma, Urine, tissue homogenates and Cell culture supernates and Other biological fluids.

Principle of the Assay||The microtiter plate provided in this kit has been pre-coated with an antibody specific to PDCD1 ligand 1. Standards or samples are then added to the appropriate microtiter plate wells with a biotin-conjugated polyclonal antibody preparation specific for PDCD1 ligand 1 and Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. Then a TMB substrate solution is added to each well. Only those wells that contain PDCD1 ligand 1, biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of a sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450 nm +/- 2 nm. The concentration of PDCD1 ligand 1 in the samples is then determined by comparing the O.D. of the samples to the standard curve.

NCBI Description

This gene encodes an immune inhibitory receptor ligand that is expressed by hematopoietic and non-hematopoietic cells, such as T cells and B cells and various types of tumor cells. The encoded protein is a type I transmembrane protein that has immunoglobulin V-like and C-like domains. Interaction of this ligand with its receptor inhibits T-cell activation and cytokine production. During infection or inflammation of normal tissue, this interaction is important for preventing autoimmunity by maintaining homeostasis of the immune response. In tumor microenvironments, this interaction provides an immune escape for tumor cells through cytotoxic T-cell inactivation. Expression of this gene in tumor cells is considered to be prognostic in many types of human malignancies, including colon cancer and renal cell carcinoma.
