Intended Uses: This CCND1 ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Porcine CCND1. This ELISA kit for research use only!

Principle of the Assay: CCND1 ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-CCND1 antibody and an CCND1-HRP conjugate. The assay sample and buffer are incubated together with CCND1-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the CCND1 concentration since CCND1 from samples and CCND1-HRP conjugate compete for the anti-CCND1 antibody binding site. Since the number of sites is limited, as more sites are occupied by CCND1 from the sample, fewer sites are left to bind CCND1-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The CCND1 concentration in each sample is interpolated from this standard curve.

NCBI Description

The protein encoded by this gene belongs to the highly conserved cyclin family, whose members are characterized by a dramatic periodicity in protein abundance throughout the cell cycle. Cyclins function as regulators of CDK kinases. Different cyclins exhibit distinct expression and degradation patterns which contribute to the temporal coordination of each mitotic event. This cyclin forms a complex with and functions as a regulatory subunit of CDK4 or CDK6, whose activity is required for cell cycle G1/S transition. This protein has been shown to interact with tumor suppressor protein Rb and the expression of this gene is regulated positively by Rb. Mutations, amplification and overexpression of this gene, which alters cell cycle progression, are observed frequently in a variety of tumors and may contribute to tumorigenesis.
