Intended Uses: This CCNA2 ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Rat CCNA2. This ELISA kit for research use only, not for therapeutic or test applications!

Principle of the Assay: CCNA2 ELISA kit applies the competitive enzyme immunoassay technique utilizing a polyclonal anti-CCNA2 antibody and an CCNA2-HRP conjugate. The assay sample and buffer are incubated together with CCNA2-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the CCNA2 concentration since CCNA2 from samples and CCNA2-HRP conjugate compete for the anti-CCNA2 antibody binding site. Since the number of sites is limited, as more sites are occupied by CCNA2 from the sample, fewer sites are left to bind CCNA2-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The CCNA2 concentration in each sample is interpolated from this standard curve.

NCBI Description

The protein encoded by this gene belongs to the highly conserved cyclin family, whose members are characterized by a dramatic periodicity in protein abundance through the cell cycle. Cyclins function as regulators of CDK kinases. Different cyclins exhibit distinct expression and degradation patterns which contribute to the temporal coordination of each mitotic event. In contrast to cyclin A1, which is present only in germ cells, this cyclin is expressed in all tissues tested. This cyclin binds and activates CDC2 or CDK2 kinases, and thus promotes both cell cycle G1/S and G2/M transitions. [provided by RefSeq, Jul 2008]
