This assay has high sensitivity and excellent specificity for detection of CCL8. No significant cross-reactivity or interference between CCL8 and analogues was observed. NOTE: Limited by current skills and knowledge, it is impossible for us to complete the cross-reactivity detection between CCL8 and all the analogues, therefore, cross reaction may still exist in some cases.

Intended Uses: This CCL8 ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Porcine CCL8. This ELISA kit for research use only, not for therapeutic or test applications!

Principle of the Assay: CCL8 ELISA kit applies the competitive enzyme immunoassay technique utilizing an anti-CCL8 antibody and an CCL8-HRP conjugate. The assay sample and buffer are incubated together with CCL8-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the CCL8 concentration since CCL8 from samples and CCL8-HRP conjugate compete for the anti-CCL8 antibody binding site. Since the number of sites is limited, as more sites are occupied by CCL8 from the sample, fewer sites are left to bind CCL8-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The CCL8 concentration in each sample is interpolated from this standard curve.

NCBI Description

This antimicrobial gene is one of several chemokine genes clustered on the q-arm of chromosome 17. Chemokines form a superfamily of secreted proteins involved in immunoregulatory and inflammatory processes. The superfamily is divided into four subfamilies based on the arrangement of N-terminal cysteine residues of the mature peptide. This chemokine is a member of the CC subfamily which is characterized by two adjacent cysteine residues. This cytokine displays chemotactic activity for monocytes, lymphocytes, basophils and eosinophils. By recruiting leukocytes to sites of inflammation this cytokine may contribute to tumor-associated leukocyte infiltration and to the antiviral state against HIV infection. The processed form MCP-2(6-76) does not show monocyte chemotactic activity, but inhibits the chemotactic effect most predominantly of CCL7, and also of CCL2 and CCL5 and CCL8. Belongs to the intercrine beta (chemokine CC) family.
