Reactivity
Canine
Specificity
This assay has high sensitivity and excellent specificity for detection of THBS2. No significant cross-reactivity or interference between THBS2 and analogues was observed. NOTE: Limited by current skills and knowledge, it is impossible for us to complete the cross-reactivity detection between THBS2 and all the analogues, therefore, cross reaction may still exist in some cases.
Samples
Serum, plasma, cell culture supernatants, body fluid and tissue homogenate
Assay Type
Quantitative Competitive
Sensitivity
0.1 ng/mL
Preparation and Storage
Store all reagents at 2-8 degree C.

Intended Uses: This THBS2 ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Canine THBS2. This ELISA kit for research use only, not for therapeutic or test applications!

Principle of the Assay: THBS2 ELISA kit applies the competitive enzyme immunoassay technique utilizing an anti-THBS2 antibody and an THBS2-HRP conjugate. The assay sample and buffer are incubated together with THBS2-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the THBS2 concentration since THBS2 from samples and THBS2-HRP conjugate compete for the anti-THBS2 antibody binding site. Since the number of sites is limited, as more sites are occupied by THBS2 from the sample, fewer sites are left to bind THBS2-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The THBS2 concentration in each sample is interpolated from this standard curve.

NCBI Description

The protein encoded by this gene belongs to the thrombospondin family. It is a disulfide-linked homotrimeric glycoprotein that mediates cell-to-cell and cell-to-matrix interactions. This protein has been shown to function as a potent inhibitor of tumor growth and angiogenesis. Studies of the mouse counterpart suggest that this protein may modulate the cell surface properties of mesenchymal cells and be involved in cell adhesion and migration. [provided by RefSeq, Jul 2008]

Uniprot Description

THBS2: Adhesive glycoprotein that mediates cell-to-cell and cell-to-matrix interactions. Ligand for CD36 mediating antiangiogenic properties. Genetic variations in THBS2 may be a cause of susceptibility to intervertebral disk disease (IDD); also known as lumbar disk herniation (LDH). IDD is one of the most common musculo-skeletal disorders and the predominant cause of low-back pain and unilateral leg pain. Belongs to the thrombospondin family.

Chromosomal Location of Human Ortholog: 6q27

Cellular Component: extracellular region; basement membrane

Molecular Function: heparin binding; protein binding; calcium ion binding

Biological Process: positive regulation of synaptogenesis; negative regulation of angiogenesis; cell adhesion

Product Notes

The Canine THBS2 thbs2 (Catalog #AAA7234429) is an ELISA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA7234429 ELISA Kit recognizes Canine THBS2. It is sometimes possible for the material contained within the vial of "Thrombospondin-2 (THBS2), ELISA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
