Intended Uses: This ADRA2A ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Canine ADRA2A. This ELISA kit for research use only!

Principle of the Assay: ADRA2A ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-ADRA2A antibody and an ADRA2A-HRP conjugate. The assay sample and buffer are incubated together with ADRA2A-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the ADRA2A concentration since ADRA2A from samples and ADRA2A-HRP conjugate compete for the anti-ADRA2A antibody binding site. Since the number of sites is limited, as more sites are occupied by ADRA2A from the sample, fewer sites are left to bind ADRA2A-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The ADRA2A concentration in each sample is interpolated from this standard curve.

NCBI Description

Alpha-2-adrenergic receptors are members of the G protein-coupled receptor superfamily. They include 3 highly homologous subtypes: alpha2A, alpha2B, and alpha2C. These receptors have a critical role in regulating neurotransmitter release from sympathetic nerves and from adrenergic neurons in the central nervous system. Studies in mouse revealed that both the alpha2A and alpha2C subtypes were required for normal presynaptic control of transmitter release from sympathetic nerves in the heart and from central noradrenergic neurons; the alpha2A subtype inhibited transmitter release at high stimulation frequencies, whereas the alpha2C subtype modulated neurotransmission at lower levels of nerve activity. This gene encodes alpha2A subtype and it contains no introns in either its coding or untranslated sequences. [provided by RefSeq, Jul 2008]
