Canine Alpha-2-macroglobulin-P (A2MP) ELISA Kit
Principle of the Assay: A1ACT ELISA kit applies the competitive enzyme immunoassay technique utilizing a polyclonal anti-A1ACT antibody and an A1ACT-HRP conjugate. The assay sample and buffer are incubated together with A1ACT-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the A1ACT concentration since A1ACT from samples and A1ACT-HRP conjugate compete for the anti-A1ACT antibody binding site. Since the number of sites is limited, as more sites are occupied by A1ACT from the sample, fewer sites are left to bind A1ACT-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The A1ACT concentration in each sample is interpolated from this standard curve.

A2M: Is able to inhibit all four classes of proteinases by a unique 'trapping' mechanism. This protein has a peptide stretch, called the 'bait region' which contains specific cleavage sites for different proteinases. When a proteinase cleaves the bait region, a conformational change is induced in the protein which traps the proteinase. The entrapped enzyme remains active against low molecular weight substrates (activity against high molecular weight substrates is greatly reduced). Following cleavage in the bait region a thioester bond is hydrolyzed and mediates the covalent binding of the protein to the proteinase. Belongs to the protease inhibitor I39 (alpha-2- macroglobulin) family.
