Principle of the Assay: ADAMTS5 ELISA kit applies the competitive enzyme immunoassay technique utilizing a polyclonal anti-ADAMTS5 antibody and an ADAMTS5-HRP conjugate. The assay sample and buffer are incubated together with ADAMTS5-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the ADAMTS5 concentration since ADAMTS5 from samples and ADAMTS5-HRP conjugate compete for the anti-ADAMTS5 antibody binding site. Since the number of sites is limited, as more sites are occupied by ADAMTS5 from the sample, fewer sites are left to bind ADAMTS5-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The ADAMTS5 concentration in each sample is interpolated from this standard curve.

This gene encodes a member of the ADAMTS (a disintegrin and metalloproteinase with thrombospondin motifs) protein family. Members of the family share several distinct protein modules, including a propeptide region, a metalloproteinase domain, a disintegrin-like domain, and a thrombospondin type 1 (TS) motif. Individual members of this family differ in the number of C-terminal TS motifs, and some have unique C-terminal domains. The enzyme encoded by this gene contains two C-terminal TS motifs and functions as aggrecanase to cleave aggrecan, a major proteoglycan of cartilage. [provided by RefSeq, Jul 2008]
