Principle of the Assay: PFKFB2 ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-PFKFB2 antibody and an PFKFB2-HRP conjugate. The assay sample and buffer are incubated together with PFKFB2-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the PFKFB2 concentration since PFKFB2 from samples and PFKFB2-HRP conjugate compete for the anti-PFKFB2 antibody binding site. Since the number of sites is limited, as more sites are occupied by PFKFB2 from the sample, fewer sites are left to bind PFKFB2-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The PFKFB2 concentration in each sample is interpolated from this standard curve.

The protein encoded by this gene is involved in both the synthesis and degradation of fructose-2,6-bisphosphate, a regulatory molecule that controls glycolysis in eukaryotes. The encoded protein has a 6-phosphofructo-2-kinase activity that catalyzes the synthesis of fructose-2,6-bisphosphate, and a fructose-2,6-biphosphatase activity that catalyzes the degradation of fructose-2,6-bisphosphate. This protein regulates fructose-2,6-bisphosphate levels in the heart, while a related enzyme encoded by a different gene regulates fructose-2,6-bisphosphate levels in the liver and muscle.
