Introduction: The immunosuppressant medicine cyclosporin A blocks a calcium-dependent signal from the T-cell receptor (TCR) which generally results in T-cell activation. cyclosporin A binds to cyclophilin B and the complex inactivates the main signaling intermediate calcineurin. CAMLG functions in the same way as cyclosporin A. CAMLG binds to cyclophilin B and operates downstream of the TCR and upstream of calcineurin by causing an influx of calcium. CAMLG is an essential membrane protein that takes part in the calcium signal transduction pathway, connecting cyclophilin B to calcium signaling, even when cyclosporine is not present.

When bound to cyclophilin B, cyclosporin A binds and inactivates the key signaling intermediate calcineurin. The protein encoded by this gene functions similarly to cyclosporin A, binding to cyclophilin B and acting downstream of the TCR and upstream of calcineurin by causing an influx of calcium.
