Principle of the Assay: CAD ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-CAD antibody and an CAD-HRP conjugate. The assay sample and buffer are incubated together with CAD-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the CAD concentration since CAD from samples and CAD-HRP conjugate compete for the anti-CAD antibody binding site. Since the number of sites is limited, as more sites are occupied by CAD from the sample, fewer sites are left to bind CAD-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The CAD concentration in each sample is interpolated from this standard curve.

NCBI Description

Apoptosis is a cell death process that removes toxic and/or useless cells during mammalian development. The apoptotic process is accompanied by shrinkage and fragmentation of the cells and nuclei and degradation of the chromosomal DNA into nucleosomal units. DNA fragmentation factor (DFF) is a heterodimeric protein of 40-kD (DFFB) and 45-kD (DFFA) subunits. DFFA is the substrate for caspase-3 and triggers DNA fragmentation during apoptosis. DFF becomes activated when DFFA is cleaved by caspase-3. The cleaved fragments of DFFA dissociate from DFFB, the active component of DFF. DFFB has been found to trigger both DNA fragmentation and chromatin condensation during apoptosis. Alternatively spliced transcript variants encoding distinct isoforms have been found for this gene but the biological validity of some of these variants has not been determined.
