Intended Uses: This TNNT2 ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Bovine TNNT2. This ELISA kit for research use only, not for therapeutic or test applications!

Principle of the Assay: TNNT2 ELISA kit applies the competitive enzyme immunoassay technique utilizing an anti-TNNT2 antibody and an TNNT2-HRP conjugate. The assay sample and buffer are incubated together with TNNT2-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the TNNT2 concentration since TNNT2 from samples and TNNT2-HRP conjugate compete for the anti-TNNT2 antibody binding site. Since the number of sites is limited, as more sites are occupied by TNNT2 from the sample, fewer sites are left to bind TNNT2-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The TNNT2 concentration in each sample is interpolated from this standard curve.

NCBI Description

The protein encoded by this gene is the tropomyosin-binding subunit of the troponin complex, which is located on the thin filament of striated muscles and regulates muscle contraction in response to alterations in intracellular calcium ion concentration. Mutations in this gene have been associated with familial hypertrophic cardiomyopathy as well as with dilated cardiomyopathy. Transcripts for this gene undergo alternative splicing that results in many tissue-specific isoforms, however, the full-length nature of some of these variants has not yet been determined. TNNT2: Troponin T is the tropomyosin-binding subunit of troponin, the thin filament regulatory complex which confers calcium-sensitivity to striated muscle actomyosin ATPase activity. Heart. The fetal heart shows a greater expression in the atrium than in the ventricle, while the adult heart shows a greater expression in the ventricle than in the atrium. Isoform 6 predominates in normal adult heart. Isoforms 1, 7 and 8 are expressed in fetal heart. Isoform 7 is also expressed in failing adult heart. Belongs to the troponin T family. 11 isoforms of the human protein are produced by alternative splicing.
