Principle of the Assay: ULK3 ELISA kit applies the competitive enzyme immunoassay technique utilizing an anti-ULK3 antibody and an ULK3-HRP conjugate. The assay sample and buffer are incubated together with ULK3-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the ULK3 concentration since ULK3 from samples and ULK3-HRP conjugate compete for the anti-ULK3 antibody binding site. Since the number of sites is limited, as more sites are occupied by ULK3 from the sample, fewer sites are left to bind ULK3-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The ULK3 concentration in each sample is interpolated from this standard curve.

ULK3: Serine/threonine protein kinase that acts as a regulator of Sonic hedgehog (SHH) signaling and autophagy. Acts as a negative regulator of SHH signaling in the absence of SHH ligand: interacts with SUFU, thereby inactivating the protein kinase activity and preventing phosphorylation of GLI proteins (GLI1, GLI2 and/or GLI3). Positively regulates SHH signaling in the presence of SHH: dissociates from SUFU, autophosphorylates and mediates phosphorylation of GLI2, activating it and promoting its nuclear translocation. Phosphorylates in vitro GLI2, as well as GLI1 and GLI3, although less efficiently. Also acts as a regulator of autophagy: following cellular senescence, able to induce autophagy. Belongs to the protein kinase superfamily. Ser/Thr protein kinase family. APG1/unc-51/ULK1 subfamily. 3 isoforms of the human protein are produced by alternative splicing.
