Intended Uses: This MFGE8 ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Bovine MFGE8. This ELISA kit for research use only, not for therapeutic or test applications!

Principle of the Assay: MFGE8 ELISA kit applies the competitive enzyme immunoassay technique utilizing a polyclonal anti-MFGE8 antibody and an MFGE8-HRP conjugate. The assay sample and buffer are incubated together with MFGE8-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the MFGE8 concentration since MFGE8 from samples and MFGE8-HRP conjugate compete for the anti-MFGE8 antibody binding site. Since the number of sites is limited, as more sites are occupied by MFGE8 from the sample, fewer sites are left to bind MFGE8-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The MFGE8 concentration in each sample is interpolated from this standard curve.

NCBI Description

This gene encodes a preproprotein that is proteolytically processed to form multiple protein products. The major encoded protein product, lactadherin, is a membrane glycoprotein that promotes phagocytosis of apoptotic cells. This protein has also been implicated in wound healing, autoimmune disease, and cancer. Lactadherin can be further processed to form a smaller cleavage product, medin, which comprises the major protein component of aortic medial amyloid (AMA). Alternative splicing results in multiple transcript variants. [provided by RefSeq, Jul 2015]

Also binds to phosphatidylserine-enriched cell surfaces in a receptor-independent manner. Zona pellucida-binding protein which may play a role in gamete interaction. Binds specifically to rotavirus and inhibits its replication. 3 isoforms of the human protein are produced by alternative splicing.
