Specificity
This assay has high sensitivity and excellent specificity for detection of MX1. No significant cross-reactivity or interference between MX1 and analogues was observed. NOTE: Limited by current skills and knowledge, it is impossible for us to complete the cross-reactivity detection between MX1 and all the analogues, therefore, cross reaction may still exist in some cases.

Intended Uses: This MX1 ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Bovine MX1. This ELISA kit for research use only, not for therapeutic or test applications!

Principle of the Assay: MX1 ELISA kit applies the competitive enzyme immunoassay technique utilizing a polyclonal anti-MX1 antibody and an MX1-HRP conjugate. The assay sample and buffer are incubated together with MX1-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the MX1 concentration since MX1 from samples and MX1-HRP conjugate compete for the anti-MX1 antibody binding site. Since the number of sites is limited, as more sites are occupied by MX1 from the sample, fewer sites are left to bind MX1-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The MX1 concentration in each sample is interpolated from this standard curve.

NCBI Description

This gene encodes a guanosine triphosphate (GTP)-metabolizing protein that participates in the cellular antiviral response. The encoded protein is induced by type I and type II interferons and antagonizes the replication process of several different RNA and DNA viruses. There is a related gene located adjacent to this gene on chromosome 21, and there are multiple pseudogenes located in a cluster on chromosome 4. Alternative splicing results in multiple transcript variants. [provided by RefSeq, Sep 2013]

Uniprot Description

MX1: Dynamin-like GTPase acting as a cell-autonomous host restriction factor against many viral pathogens. Shows activity against influenza viruses, rhabdovirus VSV, bunyavirus LACV, Thogoto virus, measles virus, Hantaan virus, Coxsackie virus CVB, Rift Valley fever virus RVFV, hepatitis B virus HBV, and Crimean- Congo hemorrhagic fever virus CCHFV. Belongs to the dynamin family. 2 isoforms of the human protein are produced by alternative splicing.

Product Notes

The Bovine MX1 mx1 (Catalog #AAA7218016) is an ELISA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA7218016 ELISA Kit recognizes Bovine MX1. It is sometimes possible for the material contained within the vial of "Interferon-induced GTP-binding protein Mx1 (MX1), ELISA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
