INTENDED USE This BECN-1 ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Bovine BECN-1. This ELISA kit for research use only!
PRINCIPLE OF THE ASSAY BECN-1 ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-BECN-1 antibody and an BECN-1-HRP conjugate. The assay sample and buffer are incubated together with BECN-1-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the BECN-1 concentration since BECN-1 from samples and BECN-1-HRP conjugate compete for the anti-BECN-1 antibody binding site. Since the number of sites is limited, as more sites are occupied by BECN-1 from the sample, fewer sites are left to bind BECN-1-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The BECN-1 concentration in each sample is interpolated from this standard curve.
NCBI and Uniprot Product Information
NCBI Description
Beclin-1 participates in the regulation of autophagy and has an important role in development, tumorigenesis, and neurodegeneration (Zhong et al., 2009 [PubMed 19270693]).[supplied by OMIM, Jul 2010]
Uniprot Description
beclin 1: an essential autophagy protein that has been linked to multiple processes including tumor suppression, protection against some cardiac and neurological degenerative diseases, and lifespan extension. A ubiquitous protein of the beclin family. Is part of the class III PI3K complex that induces autophagy. Expressed at elevated levels in dendrites and cell bodies of cerebellar Purkinje cells. Interacts with the anti-apoptotic protein Bcl-2, which inhibits autophagy when it is present in the endoplasmic reticulum. The dissociation of ATG6 from Bcl-2 is essential for its autophagic activity. Interacts with PITC and GluR-delta2. Probably forms a complex with AMBRA1 and PIK3C3. Plays a role in antiviral host defense. Protects against infection by a neurovirulent strain of Sindbis virus.

Product Notes
The Bovine Beclin 1 becn1 (Catalog #AAA742858) is an ELISA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA742858 ELISA Kit recognizes Bovine Beclin 1. It is sometimes possible for the material contained within the vial of "Beclin 1, ELISA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.Precautions
