SCNN1A blocking peptide

SCNN1A Antibody (Center) Blocking Peptide

Gene Names
SCNN1A; BESC2; ENaCa; SCNEA; SCNN1; ENaCalpha
Synonyms
SCNN1A; SCNN1A Antibody (Center) Blocking Peptide; Amiloride-sensitive sodium channel subunit alpha; Alpha-NaCH; Epithelial Na(+) channel subunit alpha; Alpha-ENaC; ENaCA; Nonvoltage-gated sodium channel 1 subunit alpha; SCNEA; SCNN1; SCNN1A blocking peptide
Ordering
Specificity
The synthetic peptide sequence used to generate the antibody was selected from the Center region of human SCNN1A. A 10 to 100 fold molar excess to antibody is recommended. Precise conditions should be optimized for a particular assay.
Form/Format
Synthetic peptide was lyophilized with 100% acetonitrile and is supplied as a powder. Reconstitute with 0.1 ml DI water for a final concentration of 1 mg/ml.
Sequence Length
669
Cellular Location
Apical cell membrane; Multi-pass membrane protein. Cell projection, cilium. Note: In the oviduct and bronchus, located on cilia in multi-ciliated cells. In endometrial non-ciliated epithelial cells, restricted to apical surfaces
Tissue Location
Expressed in the female reproductive tract, from the fimbrial end of the fallopian tube to the endometrium (at protein level). Expressed in kidney (at protein level). In the respiratory tract, expressed in the bronchial epithelium (at protein level). Highly expressed in lung. Detected at intermediate levels in pancreas and liver, and at low levels in heart and placenta. Isoform 1 and isoform 2 predominate in all tissues Expression of isoform 3, isoform 4 and isoform 5 is very low or not detectable, except in lung and heart
Preparation and Storage
Maintain refrigerated at 2-8 degree C for up to 6 months. For long term storage store at -20 degree C.

NCBI Description

Nonvoltage-gated, amiloride-sensitive, sodium channels control fluid and electrolyte transport across epithelia in many organs. These channels are heteromeric complexes consisting of 3 subunits: alpha, beta, and gamma. This gene encodes the alpha subunit, and mutations in this gene have been associated with pseudohypoaldosteronism type 1 (PHA1), a rare salt wasting disease resulting from target organ unresponsiveness to mineralocorticoids. Alternatively spliced transcript variants encoding different isoforms have been described for this gene. [provided by RefSeq, Apr 2009]

Uniprot Description

ENaC-alpha: Sodium permeable non-voltage-sensitive ion channel inhibited by the diuretic amiloride. Mediates the electrodiffusion of the luminal sodium (and water, which follows osmotically) through the apical membrane of epithelial cells. Controls the reabsorption of sodium in kidney, colon, lung and sweat glands. Also plays a role in taste perception. Probable heterotrimer containing one alpha, one beta and one gamma subunit. A delta subunit can replace the alpha subunit. Interacts with the WW domains of NEDD4, NEDD4L, WWP1 and WWP2. By aldosterone. Highly expressed in kidney and lung. Detected at intermediate levels in pancreas and liver, and at low levels in heart and placenta. Isoform 1 and isoform 2 predominate in all tissues. Expression of isoform 3, isoform 4 and isoform 5 is very low or not detectable, except in lung and heart. Activated by WNK1, WNK2, WNK3 and WNK4. Belongs to the amiloride-sensitive sodium channel (TC 1.A.6) family. SCNN1A subfamily. 6 isoforms of the human protein are produced by alternative splicing.

Protein type: Transporter, ion channel; Membrane protein, multi-pass; Channel, sodium; Membrane protein, integral

Chromosomal Location of Human Ortholog: 12p13

Cellular Component: apical plasma membrane; integral to plasma membrane; plasma membrane

Molecular Function: amiloride-sensitive sodium channel activity; protein binding; WW domain binding

Biological Process: multicellular organismal water homeostasis; sodium ion homeostasis

Disease: Bronchiectasis With Or Without Elevated Sweat Chloride 2; Pseudohypoaldosteronism, Type I, Autosomal Recessive

Product Notes

The SCNN1A scnn1a (Catalog #AAA9227859) is a Blocking Peptide and is intended for research purposes only. The product is available for immediate purchase. It is sometimes possible for the material contained within the vial of "SCNN1A, Blocking Peptide" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
