Principle of the assay: Bcl10 ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-Bcl10 antibody and an Bcl10-HRP conjugate. The assay sample and buffer are incubated together with Bcl10-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the Bcl10 concentration since Bcl10 from samples and Bcl10-HRP conjugate compete for the anti-Bcl10 antibody binding site. Since the number of sites is limited, as more sites are occupied by Bcl10 from the sample, fewer sites are left to bind Bcl10-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The Bcl10 concentration in each sample is interpolated from this standard curve.

NCBI Description

This gene was identified by its translocation in a case of mucosa-associated lymphoid tissue (MALT) lymphoma. The protein encoded by this gene contains a caspase recruitment domain (CARD), and has been shown to induce apoptosis and to activate NF-kappaB. This protein is reported to interact with other CARD domain containing proteins including CARD9, 10, 11 and 14, which are thought to function as upstream regulators in NF-kappaB signaling. This protein is found to form a complex with MALT1, a protein encoded by another gene known to be translocated in MALT lymphoma. MALT1 and this protein are thought to synergize in the activation of NF-kappaB, and the deregulation of either of them may contribute to the same pathogenetic process that leads to the malignancy. Defects in BCL10 are involved in various types of cancer.
