Specificity
This assay has high sensitivity and excellent specificity for detection of ApoB100. No significant cross-reactivity or interference between ApoB100 and analogues was observed. NOTE: Limited by current skills and knowledge, it is impossible for us to complete the cross-reactivity detection between ApoB100 and all the analogues, therefore, cross reaction may still exist in some cases.

Intended Uses: This ApoB100 ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Rat ApoB100. This ELISA kit for research use only, not for therapeutic or test applications!

Principle of the Assay: ApoB100 ELISA kit applies the competitive enzyme immunoassay technique utilizing a polyclonal anti-ApoB100 antibody and an ApoB100-HRP conjugate. The assay sample and buffer are incubated together with ApoB100-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the ApoB100 concentration since ApoB100 from samples and ApoB100-HRP conjugate compete for the anti-ApoB100 antibody binding site. Since the number of sites is limited, as more sites are occupied by ApoB100 from the sample, fewer sites are left to bind ApoB100-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The ApoB100 concentration in each sample is interpolated from this standard curve.

NCBI Description

This gene product is the main apolipoprotein of chylomicrons and low density lipoproteins. It occurs in plasma as two main isoforms, apoB-48 and apoB-100: the former is synthesized exclusively in the gut and the latter in the liver. The intestinal and the hepatic forms of apoB are encoded by a single gene from a single, very long mRNA. The two isoforms share a common N-terminal sequence. The shorter apoB-48 protein is produced after RNA editing of the apoB-100 transcript at residue 2180 (CAA->UAA), resulting in the creation of a stop codon, and early translation termination. Mutations in this gene or its regulatory region cause hypobetalipoproteinemia, normotriglyceridemic hypobetalipoproteinemia, and hypercholesterolemia due to ligand-defective apoB, diseases affecting plasma cholesterol and apoB levels. Uniprot Description

APOB: Apolipoprotein B is a major protein constituent of chylomicrons (apo B-48), LDL (apo B-100) and VLDL (apo B-100). Apo B-100 functions as a recognition signal for the cellular binding and internalization of LDL particles by the apoB/E receptor. Defects in APOB are a cause of familial hypobetalipoproteinemia type 1 (FHBL1). A disorder characterized by highly reduced plasma concentrations of low density lipoproteins, and dietary fat malabsorption. Clinical presentation may vary from no symptoms to severe gastrointestinal and neurological dysfunction similar to abetalipoproteinemia. Defects in APOB are a cause of familial ligand-defective apolipoprotein B-100 (FDB). FDB is a dominantly inherited disorder of lipoprotein metabolism leading to hypercholesterolemia and increased proneness to coronary artery disease (CAD). The plasma cholesterol levels are dramatically elevated due to impaired clearance of LDL particles by defective APOB/E receptors. Defects in APOB associated with defects in other genes (polygenic) can contribute to hypocholesterolemia.

Product Notes

The Rat Apo-B100 apob (Catalog #AAA723231) is an ELISA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA723231 ELISA Kit recognizes Rat Apo-B100. It is sometimes possible for the material contained within the vial of "Apoprotein B100, ELISA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
