Principle of the assay: ANXA7 ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-ANXA7 antibody and an ANXA7-HRP conjugate. The assay sample and buffer are incubated together with ANXA7-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the ANXA7 concentration since ANXA7 from samples and ANXA7-HRP conjugate compete for the anti-ANXA7 antibody binding site. Since the number of sites is limited, as more sites are occupied by ANXA7 from the sample, fewer sites are left to bind ANXA7-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The ANXA7 concentration in each sample is interpolated from this standard curve.

NCBI Description

Annexin VII is a member of the annexin family of calcium-dependent phospholipid binding proteins. The Annexin VII gene contains 14 exons and spans approximately 34 kb of DNA. An alternatively spliced cassette exon results in two mRNA transcripts of 2.0 and 2.4 kb which are predicted to generate two protein isoforms differing in their N-terminal domain. The alternative splicing event is tissue specific and the mRNA containing the cassette exon is prevalent in brain, heart and skeletal muscle. The transcripts also differ in their 3'-non coding regions by the use of two alternative poly(A) signals. Annexin VII encodes a protein with a molecular weight of approximately 51 kDa with a unique, highly hydrophobic N-terminal domain of 167 amino acids and a conserved C-terminal region of 299 amino acids. The latter domain is composed of alternating hydrophobic and hydrophilic segments. Structural analysis of the protein suggests that Annexin VII is a membrane binding protein with diverse properties, including voltage-sensitive calcium channel activity, ion selectivity and membrane fusion. Uniprot Description

ANXA7: Calcium/phospholipid-binding protein which promotes membrane fusion and is involved in exocytosis. Interacts with PDCD6. Isoform 1 is expressed in brain, heart and skeletal muscle. Isoform 2 is more abundant in liver, lung, kidney, spleen, fibroblasts and placenta. Belongs to the annexin family. 2 isoforms of the human protein are produced by alternative splicing.

Product Notes

The Chicken ANXA7 anxa7 (Catalog #AAA747186) is an ELISA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA747186 ELISA Kit recognizes Chicken ANXA7. It is sometimes possible for the material contained within the vial of "Annexin A7, ELISA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
