Principle of the assay: alpha2MG ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-alpha2MG antibody and an alpha2MG-HRP conjugate. The assay sample and buffer are incubated together with alpha2MG-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the alpha2MG concentration since alpha2MG from samples and alpha2MG-HRP conjugate compete for the anti-alpha2MG antibody binding site. Since the number of sites is limited, as more sites are occupied by alpha2MG from the sample, fewer sites are left to bind alpha2MG-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The alpha2MG concentration in each sample is interpolated from this standard curve.

NCBI Description

Alpha-2-macroglobulin is a protease inhibitor and cytokine transporter. It inhibits many proteases, including trypsin, thrombin and collagenase. A2M is implicated in Alzheimer disease (AD) due to its ability to mediate the clearance and degradation of A-beta, the major component of beta-amyloid deposits. [provided by RefSeq, Jul 2008]

Uniprot Description

A2M: Is able to inhibit all four classes of proteinases by a unique 'trapping' mechanism. This protein has a peptide stretch, called the 'bait region' which contains specific cleavage sites for different proteinases. When a proteinase cleaves the bait region, a conformational change is induced in the protein which traps the proteinase. The entrapped enzyme remains active against low molecular weight substrates (activity against high molecular weight substrates is greatly reduced). Following cleavage in the bait region a thioester bond is hydrolyzed and mediates the covalent binding of the protein to the proteinase. Belongs to the protease inhibitor I39 (alpha-2- macroglobulin) family.
