Principle of the Assay: ALDOA ELISA kit applies the competitive enzyme immunoassay technique utilizing a polyclonal anti-ALDOA antibody and an ALDOA-HRP conjugate. The assay sample and buffer are incubated together with ALDOA-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the ALDOA concentration since ALDOA from samples and ALDOA-HRP conjugate compete for the anti-ALDOA antibody binding site. Since the number of sites is limited, as more sites are occupied by ALDOA from the sample, fewer sites are left to bind ALDOA-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The ALDOA concentration in each sample is interpolated from this standard curve.

The protein encoded by this gene, Aldolase A (fructose-bisphosphate aldolase), is a glycolytic enzyme that catalyzes the reversible conversion of fructose-1,6-bisphosphate to glyceraldehyde 3-phosphate and dihydroxyacetone phosphate. Three aldolase isozymes (A, B, and C), encoded by three different genes, are differentially expressed during development. Aldolase A is found in the developing embryo and is produced in even greater amounts in adult muscle. Aldolase A expression is repressed in adult liver, kidney and intestine and similar to aldolase C levels in brain and other nervous tissue. Aldolase A deficiency has been associated with myopathy and hemolytic anemia. Alternative splicing and alternative promoter usage results in multiple transcript variants.
