Principle of the assay: ADAMTS4 ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-ADAMTS4 antibody and an ADAMTS4-HRP conjugate. The assay sample and buffer are incubated together with ADAMTS4-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the ADAMTS4 concentration since ADAMTS4 from samples and ADAMTS4-HRP conjugate compete for the anti-ADAMTS4 antibody binding site. Since the number of sites is limited, as more sites are occupied by ADAMTS4 from the sample, fewer sites are left to bind ADAMTS4-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The ADAMTS4 concentration in each sample is interpolated from this standard curve.

NCBI Description

This gene encodes a member of the ADAMTS (a disintegrin and metalloproteinase with thrombospondin motifs) protein family. Members of the family share several distinct protein modules, including a propeptide region, a metalloproteinase domain, a disintegrin-like domain, and a thrombospondin type 1 (TS) motif. Individual members of this family differ in the number of C-terminal TS motifs, and some have unique C-terminal domains. The enzyme encoded by this gene lacks a C-terminal TS motif. It is responsible for the degradation of aggrecan, a major proteoglycan of cartilage, and brevican, a brain-specific extracellular matrix protein. The cleavage of aggrecan and brevican suggests key roles of this enzyme in arthritic disease and in the central nervous system, potentially, in the progression of glioma.
