Assay Type
Sandwich
Detection Range
15.6pg/ml-500pg/ml.
Sensitivity
2.0pg/ml.
Intended Uses
This Quantitative Sandwich ELISA kit is only for in vitro research use only, not for drug, household, therapeutic applications! It is intended to be determinated ADAM10 concentrations in Cat serum, plasma and other body fluids. Using Purified Cat ADAM10 antibody to coat Microelisa Stripplate wells to make solid-phase antibody, then add ADAM10 and ADAM10 antibody which has been labeled with HRP to wells, then the reactants become antibody-antigen-antibody-enzyme complex, after washing completely, add TMB substrate solution, TMB substrate becomes blue color under HRP enzyme-catalyzed, reaction is terminated by the addition of a sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450 nm. The concentration of ADAM10 in the samples is then determined by comparing the O.D. of the samples to the standard curve.
Intra-assay Precision
Intra-assay CV (%) is less than 15%.
Inter-assay Precision
Inter-assay CV (%) is less than 15%. [CV(%) = SD/mean ×100]
Preparation and Storage
Store all reagents at 2-8 degree C

NCBI Description

Members of the ADAM family are cell surface proteins with a unique structure possessing both potential adhesion and protease domains. This gene encodes and ADAM family member that cleaves many proteins including TNF-alpha and E-cadherin. [provided by RefSeq, Jul 2008]

Uniprot Description

ADAM10: Cleaves the membrane-bound precursor of TNF-alpha at '76-Ala-|-Val-77' to its mature soluble form. Responsible for the proteolytical release of soluble JAM3 from endothelial cells surface. Responsible for the proteolytic release of several other cell-surface proteins, including heparin-binding epidermal growth- like factor, ephrin-A2 and for constitutive and regulated alpha- secretase cleavage of amyloid precursor protein (APP). Contributes to the normal cleavage of the cellular prion protein. Involved in the cleavage of the adhesion molecule L1 at the cell surface and in released membrane vesicles, suggesting a vesicle-based protease activity. Controls also the proteolytic processing of Notch and mediates lateral inhibition during neurogenesis. Responsible for the FasL ectodomain shedding and for the generation of the remnant ADAM10-processed FasL (FasL APL) transmembrane form. Also cleaves the ectodomain of the integral membrane proteins CORIN and ITM2B. May regulate the EFNA5-EPHA3 signaling.

Product Notes

The Cat ADAM10 adam10 (Catalog #AAA076818) is an ELISA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA076818 ELISA Kit recognizes Cat ADAM10. It is sometimes possible for the material contained within the vial of "A Disintegrin and Metalloprotease 10, ELISA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
