• Sequence: The sequence of the first five N-terminal amino acids was determined and was found to be Asn-Ser-Asp-Ser-Glu, which agrees with the sequence of native human EGF. N-terminal methionine has been completely removed enzymatically.

• Biological Activity: Recombinant Human EGF is fully biologically active when compared to standards. The ED50, calculated by the dose-dependent proliferation of murine BALB/c 3T3 cells (measured by 3H-thymidine uptake) <0.1ng/ml corresponding to a specific activity of 1x10e7 U/mg.
• Protein Content: Protein quantitation was carried out by two independent methods:; 1. UV spectroscopy at 280nm using the absorbency value of 2.858 as the extinction coefficient for a 0.1% (1mg/ml) solution. This value is calculated by the PC GENE computer analysis program of protein sequences (IntelliGenetics).; 2. Analysis by RP-HPLC, using a calibrated solution of human EGF as a Reference Standard.
• Preparation and Storage: Lyophilized powder may be stored at -20 degree C. Stable for 12 months at -20 degree C. Reconstitute with sterile dH2O, 0.1% HSA or BSA. Aliquot to avoid repeated freezing and thawing. Store at -20 degree C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

NCBI Description

This gene encodes a member of the epidermal growth factor superfamily. The encoded protein is synthesized as a large precursor molecule that is proteolytically cleaved to generate the 53-amino acid epidermal growth factor peptide. This protein acts a potent mitogenic factor that plays an important role in the growth, proliferation and differentiation of numerous cell types. This protein acts by binding the high affinity cell surface receptor, epidermal growth factor receptor. Defects in this gene are the cause of hypomagnesemia type 4. Dysregulation of this gene has been associated with the growth and progression of certain cancers. Alternate splicing results in multiple transcript variants.
