Unit Definition
One unit of the recombinant caspase-7 is the enzyme activity that cleaves 1nmol of the caspase substrate DEVD-pNA (pNA: pnitroanaline) per hour at 37 degree C in a reaction solution containing 50mM Hepes, pH 7.2, 50mM NaCl, 0.1% CHAPS, 10mM EDTA, 5% glycerol, and 10mM DTT.
Preparation and Storage
Lyophilized powder may be stored at -70 degree C. Reconstitute with sterile 15% glycerol, PBS. Aliquot and store at -70 degree C. Reconstituted product is stable for 6 months at -70 degree C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

Caspase-7 (also know as Mch3, ICE-LAP3, CMH-1) is a member of the interleukin-1b converting enzyme (ICE) family of cysteine proteases. Same as other caspases, caspase-7 also exists in cells as an inactive proenzyme. During apoptosis procaspase-7 is processed at aspartate residues by self-proteolysis and/or cleavage by another caspase. The processed form of caspase-7 consists of large and small subunits which associate to form the active enzyme. The active caspase-7 has been shown involving in the proteolysis of poly (ADP-ribose) polymerase (PARP), an enzyme that is involved in DNA repair and genomic maintenance. The upstream sequence of the PARP cleavage site, DEVD, has been utilized as a basis for synthesis of caspase-3 and -7 substrates DEVD-AFC and DEVD-pNA. The active recombinant caspase-7 is routinely tested for its ability to enzymatically cleave the caspase-7 colorimetric substrate Ac-DEVD-pNA.

The active recombinant human caspase-7 was expressed in E. coli. The expressed caspase-7 spontaneously undergoes autoprocessing to yield the subunits characteristic of the native enzyme. The recombinant caspase-7 preferentially cleaves the substrates consisting of consensus sequence DEVD. In combination with caspase activity assays, the active caspase-7 is useful in screening caspase inhibitors, studying enzyme regulation, determining specificities of caspase substrates, as well as using as a positive control in caspase activity assays and Western blot analyses. We recommend using 1 unit per assay for analyzing caspase activity.
