Unit Definition
One unit of the recombinant caspase-2 is the enzyme activity that cleaves 1 nmol of the caspase substrate VDVAD-pNA (pNA: pnitroanaline) per hour at 37 degree C in a reaction solution containing 50mM Hepes, pH 7.2, 50mM NaCl, 0.1% Chaps, 10mM EDTA, 5% Glycerol, and 10mM DTT.
Preparation and Storage
The lyophilized caspase-2 is stable for 1 year at -70 degree C. Following reconstitution in PBS, the enzyme should be aliquoted and stored at -70 degree C. Avoid multiple freeze/thaw cycles as activity might decrease.

Recombinant Human Caspases

MyBioSource offers the most comprehensive list of highly active caspases. The recombinant forms of active caspases were expressed in E. coli and puri ed, most with purity >90% by SDS-PAGE. Active caspases are very useful in studying enzyme regulation and kinetics, screening caspase inhibitors, determining target substrates, and as positive controls in caspase activity assays and Western blot analyses, etc. One unit of each active enzyme is suf cient to cleave 1 nmol of the corresponding caspase substrate per hour at 37 degree C. Procaspase-3 are also available for studying caspase activation pathways and mechanisms.
Caspase-2 (also know as Ich-1, Nedd-2) is a member of the interleukin-1b converting enzyme (ICE) family of cysteine proteases. Similar as other caspases, caspase-2 also exists in cells as an inactive proenzyme. During apoptosis, procaspase-2 is processed at aspartate residues by self-proteolysis and/or cleavage by upstream caspases. The processed form of caspase-2 consists of large (19kD) and small (12kD) subunits which associate to form the active enzyme. The active recombinant human caspase-2 was expressed in E. coli. The expressed caspase-2 spontaneously undergoes auto-processing to yield the subunits characteristic of the native enzyme. The active recombinant caspase-2 is routinely tested for its ability to enzymatically cleave the caspase-2 colorimetric substrate Ac-VDVAD-pNA. The signal can be detected by spectrophotometry.
