Form/Format
Supplied as a lyophilized powder from 30% Acetonitrile, 0.1% TFA, 1.25mg BSA. Reconstitute to 10ug/ml in sterile 4mM HCI, with 0.1% HSA or BSA.
Activity
Measured by its ability to induce alkaline phosphatase synthesis by MC3T3E1 osteoblastic cells (Mazerbourg, S. et al., 2005, J. Biol. Chem. 280:32122). The ED50 for this effect is typically 15-60ng/mL.
Reconstitution
It is recommended that sterile 4mM HCI containing at least 0.1% human serum albumin or bovine serum albumin be added to the vial to prepare a stock solution of no less than 10ug/mL.
Preparation and Storage
Lyophilized powder may be stored at -20 degree C. Stable for 12 months at -20 degree C. Reconstitute with sterile 4mM HCI, with 0.1% HSA or BSA. Aliquot to avoid repeated freezing and thawing. Store at -20 degree C. Reconstituted product is stable for 6 months at -20 degree C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

The mature recombinant human BMP-10, generated by the proteolytic removal of the signal peptide and the propeptide, is a disulfide-linked homodimeric protein. Based on N-terminal sequencing, the mature recombinant human BMP-10 starts at Asn 317. The monomer has a calculated molecular mass of 12.2kD and migrates as an approximately 12.2kD protein in SDS-PAGE under reducing conditions.

The protein encoded by this gene is a member of the TGF-beta family of growth factors. Data suggest that the similar protein in mouse plays an important role in trabeculation of the embryonic heart. In human, this protein may signal through receptor serine/threonine kinases. [provided by RefSeq]

Function: Required for maintaining the proliferative activity of embryonic cardiomyocytes by preventing premature activation of the negative cell cycle regulator CDKN1C/p57KIP and maintaining the required expression levels of cardiogenic factors such as MEF2C and NKX2-5. Acts as a ligand for ACVRL1/ALK1, BMPR1A/ALK3 and BMPR1B/ALK6, leading to activation of SMAD1, SMAD5 and SMAD8 transcription factors. Inhibits endothelial cell migration and growth. Ref.4 Ref.5
