Human Actin Alpha 1, Skeletal Muscle ELISA Kit | ACTAlpha 1 elisa kit
Human Actin Alpha 1, Skeletal Muscle ELISA Kit
Intended Uses: This ACTA1 ELISA kit is intended for laboratory research use only. The stop solution changes the color from blue to yellow and the intensity of the color is measured at 450 nm using a spectrophotometer. In order to measure the concentration of ACTA1 in the sample, this ACTA1 ELISA Kit includes a set of calibration standards. The calibration standards are assayed at the same time as the samples and allow the operator to produce a standard curve of Optical Density versus ACTA1 concentration. The concentration of in the samples is then determined by comparing the O.D. of the samples to the standard curve.
Principle of the Assay: The coated well immunoenzymatic assay for the quantitative measurement of ACTA1 utilizes a multiclonal anti-ACTA1 antibody and an ACTA1-HRP conjugate. The assay sample and buffer are incubated together with ACTA1-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the ACTA1 concentration since ACTA1 from samples and ACTA1-HRP conjugate compete for the anti-ACTA1 antibody binding site. Since the number of sites is limited, as more sites are occupied by ACTA1 from the sample, fewer sites are left to bind ACTA1-HRP conjugate. Standards of known ACTA1 concentrations are run concurrently with the samples being assayed and a standard curve is plotted relating the intensity of the color (Optical Density) to the concentration of ACTA1. The ACTA1 concentration in each sample is interpolated from this standard curve.
NCBI and Uniprot Product Information
NCBI Description
The protein encoded by this gene is a plasma protease inhibitor and member of the serine protease inhibitor class. Polymorphisms in this protein appear to be tissue specific and influence protease targeting. Variations in this protein's sequence have been implicated in Alzheimer's disease, and deficiency of this protein has been associated with liver disease. Mutations have been identified in patients with Parkinson disease and chronic obstructive pulmonary disease. [provided by RefSeq, Jul 2008]
Uniprot Description
SERPINA3: Although its physiological function is unclear, it can inhibit neutrophil cathepsin G and mast cell chymase, both of which can convert angiotensin-1 to the active angiotensin-2. Belongs to the serpin family. 3 isoforms of the human protein are produced by alternative splicing.

Product Notes
The Human ACTAlpha 1 serpina3 (Catalog #AAA732582) is an ELISA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA732582 ELISA Kit recognizes Human ACTAlpha 1. It is sometimes possible for the material contained within the vial of "Actin Alpha 1, Skeletal Muscle, ELISA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.Precautions
