Intended Uses: This ACE2 ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Monkey ACE2. This ELISA kit for research use only, not for therapeutic or test applications!

Principle of the Assay: ACE2 ELISA kit applies the competitive enzyme immunoassay technique utilizing a polyclonal anti-ACE2 antibody and an ACE2-HRP conjugate. The assay sample and buffer are incubated together with ACE2-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the ACE2 concentration since ACE2 from samples and ACE2-HRP conjugate compete for the anti-ACE2 antibody binding site. Since the number of sites is limited, as more sites are occupied by ACE2 from the sample, fewer sites are left to bind ACE2-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The ACE2 concentration in each sample is interpolated from this standard curve.

NCBI Description

The protein encoded by this gene belongs to the angiotensin-converting enzyme family of dipeptidyl carboxydipeptidases and has considerable homology to human angiotensin 1 converting enzyme. This secreted protein catalyzes the cleavage of angiotensin I into angiotensin 1-9, and angiotensin II into the vasodilator angiotensin 1-7. The organ- and cell-specific expression of this gene suggests that it may play a role in the regulation of cardiovascular and renal function, as well as fertility. In addition, the encoded protein is a functional receptor for the spike glycoprotein of the human coronaviruses SARS and HCoV-NL63. [provided by RefSeq, Jul 2008]

Also able to hydrolyze apelin- 13 and dynorphin-13 with high efficiency. May be an important regulator of heart function. In case of human coronaviruses SARS and HCoV-NL63 infections, serve as functional receptor for the spike glycoprotein of both coronaviruses. Interacts with ITGB1. Interacts with SARS-CoV and HCoV- NL63 spike glycoprotein. Up-regulated in failing heart. Expressed in endothelial cells from small and large arteries, and in arterial smooth muscle cells. Expressed in lung alveolar epithelial cells, enterocytes of the small intestine, Leydig cells and Sertoli cells. Expressed in heart, kidney, testis, and gastrointestinal system. Activated by chloride and fluoride, but not bromide. Inhibited by MLN-4760, cFP_Leu, and EDTA, but not by the ACE inhibitors linosipril, captopril and enalaprilat. Belongs to the peptidase M2 family. 2 isoforms of the human protein are produced by alternative splicing.
