(Hydrolysis of recombinant aggrecan interglobular domain with isolated ADAMTS4. Recombinant aggrecan-IGD1 at a concentration of 10uM in 50mM Tris-HCl, pH7.5, 150mM NaCl, 5mM CaCl2 was incubated with out and with 0.1uM truncated ADAMTS-4 for various time intervals. Aliquots with 1.5ug aggrecan-IGD1 were then analyzed by SDS-PAGE. This gene is a member of the aggrecan/versican proteoglycan family. The encoded protein is an integral part of the extracellular matrix in cartilagenous tissue and it withstands compression in cartilage. Mutations in this gene may be involved in skeletal dysplasia and spinal degeneration. ACAN: This proteoglycan is a major component of extracellular matrix of cartilagenous tissues. A major function of this protein is to resist compression in cartilage. It binds avidly to hyaluronic acid via an N-terminal globular region. Defects in ACAN are the cause of spondyloepiphyseal dysplasia type Kimberley (SEDK). Spondyloepiphyseal dysplasias are a heterogeneous group of congenital chondrodysplasias that specifically affect epiphyses and vertebrae. The autosomal dominant SEDK is associated with premature degenerative arthropathy. Defects in ACAN are the cause of spondyloepimetaphyseal dysplasia aggrecan type (SEMD-ACAN). A bone disease characterized by severe short stature, macrocephaly, severe midface hypoplasia, short neck, barrel chest and brachydactyly. The radiological findings comprise long bones with generalized irregular epiphyses with widened metaphyses, especially at the knees, platyspondyly, and multiple cervical-vertebral clefts. Defects in ACAN are the cause of osteochondritis dissecans short stature and early-onset osteoarthritis (OD). It is a type of osteochondritis defined as a separation of cartilage and subchondral bone from the surrounding tissue, primarily affecting the knee, ankle and elbow joints. It is clinically characterized by multiple osteochondritic lesions in knees and/or hips and/or elbows, disproportionate short stature and early-onset osteoarthritis. Belongs to the aggrecan/versican proteoglycan family. 3 isoforms of the human protein are produced by alternative splicing.

The ACAN acan (Catalog #AAA637420) is a Recombinant Protein and is intended for research purposes only. The product is available for immediate purchase. AAA Biotech's Aggrecan, Interglobular Domain can be used in a range of immunoassay formats including, but not limited to, ELISA (EL/EIA). Aggrecan interglobular domain is used as substrate for aggrecanases and matrix metalloproteinases. For proteinase activity measurements, the protein is incubated with proteinase for various time intervals. Thereafter, aliquots of the incubation mixture are analyzed by SDS-PAGE or by ELISA. Upon cleavage with aggrecanases the apparent Mr of aggrecan interlobular domain in SDS-PAGE is reduced from 21kD to ~13kD. Quantitative measurement of aggrecanase cleavage requires a neoepitope antibody with specificity for the N-terminus ARGSVILT.. appearing upon hydrolysis. The fragment with the newly formed N-terminus is fixed by the neoepitope antibody to a microplate and quantified with an anti-His-tag antibody. In analogy, cleavage by matrix metalloproteinases can be measured with antibodies to neoepitopes appearing upon action of these enzymes. Researchers should empirically determine the suitability of the ACAN acan for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process.
