Usage Notes
* Do not mix components from different kit lots or use reagents beyond the kit expiration date.
* Allow all reagents to warm to room temperature for at least 30 minutes before opening.
* Pre-rinse the pipet tip with reagent, use fresh pipet tips for each sample, standard and reagent to avoid contamination.
* Unused wells must be kept desiccated at 4 degree C in the sealed bag provided.
* Mix Thoroughly is very important for the result. It is recommended using low frequency oscillator or slight hand shaking every 10 minutes.
* It is recommended that all samples and standards be assayed in duplicate or triplicate.
Preparation and Storage
Store at 2-8 degree C.

Description: This Human Beta-adrenergic receptor kinase 1 (ADRBK1) ELISA Kit employs a two-site sandwich ELISA to quantitate ADRBK1 in samples. An antibody specific for ADRBK1 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any ADRBK1 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for ADRBK1 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of ADRBK1 bound in the initial step. The color development is stopped and the intensity of the color is measured.

Background: Beta-adrenergic receptor kinase (BARK) phosphorylates the beta-2-adrenergic receptor and appears to mediate agonist-specific desensitization observed at high agonist concentrations. BARK is a ubiquitous cytosolic enzyme that specifically phosphorylates the activated form of the beta-adrenergic and related G protein-coupled receptors. By study of rodent/human hybrid cells retaining various human chromosomes and parts of chromosomes, Benovic et al. (1991) demonstrated that the ADRBK1 gene segregates with the long arm of chromosome 11, centromeric to 11q13, i.e., 11cen-q13. Benovic et al. (1991) mapped the homologous gene to mouse chromosome 19.

NCBI Description

This gene encodes a member of the G protein-coupled receptor kinase family of proteins. The encoded protein phosphorylates the beta-adrenergic receptor as well as a wide range of other substrates including non-GPCR cell surface receptors, and cytoskeletal, mitochondrial, and transcription factor proteins. Data from rodent models supports a role for this gene in embryonic development, heart function and metabolism. Elevated expression of this gene has been observed in human patients with heart failure and Alzheimer's disease. [provided by RefSeq, Sep 2017]

Mouse models of severe heart failure have been used to demonstrate that inhibition of BARK1 with a peptide inhibitor is sufficient to increase mean survival, reduce dialation and improve cardiac function.
