Usage Notes
* Do not mix components from different kit lots or use reagents beyond the kit expiration date.
* Allow all reagents to warm to room temperature for at least 30 minutes before opening.
* Pre-rinse the pipet tip with reagent, use fresh pipet tips for each sample, standard and reagent to avoid contamination.
* Unused wells must be kept desiccated at 4 degree C in the sealed bag provided.
* Mix Thoroughly is very important for the result. It is recommended using low frequency oscillator or slight hand shaking every 10 minutes.
* It is recommended that all samples and standards be assayed in duplicate or triplicate.
Preparation and Storage
Store at 2-8 degree C.

Related Product Information for SLC11A2 elisa kit
Description: This Human Natural resistance-associated macrophage protein 2 (SLC11A2) ELISA Kit employs a two-site sandwich ELISA to quantitate SLC11A2 in samples. An antibody specific for SLC11A2 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any SLC11A2 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for SLC11A2 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of SLC11A2 bound in the initial step. The color development is stopped and the intensity of the color is measured.

Background: The SLC11A2 gene encodes a divalent metal transporter (DMT1), which carries iron, manganese, cobalt, nickel, cadmium, lead, copper, and zinc. DMT1 participates in cellular iron absorption at the luminal surface of the duodenum as well as in other areas of the body. Predicted amino acid sequence analysis of the NRAMP2 polypeptide identified a polytopic membrane protein highly homologous to human NRAMP1, with 66% identical residues (80% overall homology), resulting in identical predicted secondary structure of the 2 proteins. Sequence conservation is particularly high in the predicted transmembrane domains (90%), suggesting that these regions play a key role in the structural and functional aspects common to both proteins. As opposed to its NRAMP1 counterpart, whose expression is restricted to phagocytic cells.

NCBI Description

This gene encodes a member of the solute carrier family 11 protein family. The product of this gene transports divalent metals and is involved in iron absorption. Mutations in this gene are associated with hypochromic microcytic anemia with iron overload. A related solute carrier family 11 protein gene is located on chromosome 2. Multiple transcript variants encoding different isoforms have been found for this gene.[provided by RefSeq, Apr 2010]

Uniprot Description

SLC11A2: Important in metal transport, in particular iron. Can also transport manganese, cobalt, cadmium, nickel, vanadium and lead. Involved in apical iron uptake into duodenal enterocytes. Involved in iron transport from acidified endosomes into the cytoplasm of erythroid precursor cells. May play an important role in hepatic iron accumulation and tissue iron distribution. Defects in SLC11A2 are a cause of hypochromic microcytic anemia (HCMA). The disease is characterized by an abnormal hemoglobin content in the erythrocytes which are reduced in size. It may be hereditary or acquired. Mutations in SLC11A2 are associated with progressive liver iron overload and normal to moderately elevated serum ferritin levels. Belongs to the NRAMP family. 4 isoforms of the human protein are produced by alternative splicing.

Product Notes

The Human SLC11A2 slc11a2 (Catalog #AAA9714296) is an ELISA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA9714296 ELISA Kit recognizes Human SLC11A2. It is sometimes possible for the material contained within the vial of "Natural Resistance-Associated Macrophage Protein 2 (SLC11A2), ELISA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
