Principle of the Assay: Human Taste Receptor Type 1 Member 2 (TAS1R2) ELISA Kit employs a two-site sandwich ELISA to quantitate TAS1R2 in samples. An antibody specific for TAS1R2 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any TAS1R2 present is bound by the immobilized antibody. After removing any unbound substances, HRP-Conjugated Human TAS1R2 detection antibody is added to the wells. Following a wash to remove any unbound HRP reagent, a Chromogen solution is added to the wells and color develops in proportion to the amount of TAS1R2 bound in the initial step. The color development is stopped and the intensity of the color is measured.

Background/Introduction: Taste receptor type 1 member 2 (TAS1R2) is a G protein-coupled receptor with seven trans-membrane domains and is a component of the heterodimeric amino acid taste receptor T1R2+3. TAS1R2 is formed as a dimer of the TAS1R2 and TAS1R3 proteins. Moreover, the TAS1R2 protein is not functional without formation of the 2+3 heterodimer. Another interesting quality of the TAS1R2 and TAS1R1 genes is their spontaneous activity in the absence of the extracellular domains and binding ligands. This may mean that the extracellular domain regulates function of the receptor by preventing spontaneous action as well as binding to activating ligands such as sucrose.
