Usage Notes
* Do not mix components from different kit lots or use reagents beyond the kit expiration date.
* Allow all reagents to warm to room temperature for at least 30 minutes before opening.
* Pre-rinse the pipet tip with reagent, use fresh pipet tips for each sample, standard and reagent to avoid contamination.
* Unused wells must be kept desiccated at 4 degree C in the sealed bag provided.
* Mix Thoroughly is very important for the result. It is recommended using low frequency oscillator or slight hand shaking every 10 minutes.
* It is recommended that all samples and standards be assayed in duplicate or triplicate.
Preparation and Storage
Store at 2-8 degree C.

Related Product Information for AKT2 elisa kit
Description: This Rat RAC-beta serine/threonine-protein kinase (AKT2) ELISA Kit employs a two-site sandwich ELISA to quantitate AKT2 in samples. An antibody specific for AKT2 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any AKT2 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for AKT2 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of AKT2 bound in the initial step. The color development is stopped and the intensity of the color is measured.

Background: AKT2 is a putative oncogene encoding a protein belonging to a subfamily of serine/threonine kinases containing SH2-like (Src homology 2-like) domains. The gene was shown to be amplified and overexpressed in 2 of 8 ovarian carcinoma cell lines and 2 of 15 primary ovarian tumors. Overexpression contributes to the malignant phenotype of a subset of human ductal pancreatic cancers. The encoded protein is a general protein kinase capable of phosphorylating several known proteins. Mice lacking Akt2 have a normal body mass, but display a profound diabetic phenotype, indicating that Akt2 plays a key role in signal transduction downstream of the insulin receptor. Mice lacking Akt2 show worse outcome in breast cancer initaited by the large T antigen as well as the neu oncogene.

NCBI Description

may be involved in phosphatidylinositol 3-kinase (PI3-K) mediated signaling [RGD, Feb 2006]

Uniprot Description

Akt2: an AGC kinase. Plays critical roles in glucose metabolism and the development or maintenance of proper adipose tissue and islet mass for which other Akt/PKB isoforms are unable to fully compensate. Amplified and overexpressed in human ovarian carcinoma cell lines and amplified in some primary ovarian and pancreatic tumors. Antisense blocks invasiveness in xenografts. Expressed in several insulin-responsive tissues, and one case of Type II diabetes has been associated with a likely LOF point mutation. Mouse mutants have defects in insulin response.

Product Notes

The Rat AKT2 akt2 (Catalog #AAA9711879) is an ELISA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA9711879 ELISA Kit recognizes Rat AKT2. It is sometimes possible for the material contained within the vial of "RAC-beta Serine/Threonine-Protein Kinase (AKT2), ELISA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
