Similarity
Belongs to the class I-like SAM-binding methyltransferase superfamily. rRNA adenine N(6)-methyltransferase family. KsgA subfamily.
Subunit Structure
Homodimer. Component of the mitochondrial transcription initiation complex, composed at least of TFB2M, TFAM and POLRMT. In this complex TFAM recruits POLRMT to the promoter whereas TFB2M induces structural changes in POLRMT to enable promoter opening and trapping of the DNA non-template strand. Interacts with mitochondrial RNA polymerase POLRMT. Interacts with TFAM.
Tissue Specificity
Ubiquitously expressed.
Function
S-adenosyl-L-methionine-dependent rRNA methyltransferase which may methylate two specific adjacent adenosines in the loop of a conserved hairpin near the 3'-end of 12S mitochondrial rRNA (Probable). Component of the mitochondrial transcription initiation complex, composed at least of TFB2M, TFAM and POLRMT that is required for basal transcription of mitochondrial DNA. In this complex, TFAM recruits POLRMT to a specific promoter whereas TFB2M induces structural changes in POLRMT to enable promoter opening and trapping of the DNA non-template strand. Stimulates transcription independently of the methyltransferase activity.
Preparation and Storage
Store at -20 degree C. Stable for 12 months from date of receipt.

Uniprot Description

TFB2M: S-adenosyl-L-methionine-dependent methyltransferase which specifically dimethylates mitochondrial 12S rRNA at the conserved stem loop. Also required for basal transcription of mitochondrial DNA, probably via its interaction with POLRMT and TFAM. Stimulates transcription independently of the methyltransferase activity. Compared to TFB1M, it activates transcription of mitochondrial DNA more efficiently, while it has less methyltransferase activity. Belongs to the methyltransferase superfamily. rRNA adenine N(6)-methyltransferase family. KsgA subfamily.

Protein type: EC 2.1.1.-; Methyltransferase; Transcription, coactivator/corepressor

Chromosomal Location of Human Ortholog: 1q44

Cellular Component: mitochondrial matrix

Molecular Function: rRNA (adenine-N6,N6-)-dimethyltransferase activity; transcription cofactor activity

Biological Process: mitochondrion organization and biogenesis; positive regulation of transcription, DNA-dependent; organelle organization and biogenesis; rRNA methylation; gene expression; transcription from mitochondrial promoter; transcription initiation from mitochondrial promoter

Product Notes

The TFB2M tfb2m (Catalog #AAA9629099) is an Antibody produced from Rabbit and is intended for research purposes only. The product is available for immediate purchase. The TFB2M Antibody reacts with Human, Mouse and may cross-react with other species as described in the data sheet. AAA Biotech's TFB2M can be used in a range of immunoassay formats including, but not limited to, ELISA (EIA)(peptide). ELISA(peptide): 1:20000-1:40000. Researchers should empirically determine the suitability of the TFB2M tfb2m for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "TFB2M, Polyclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
