Western Blot (WB)
(Western blot analysis of extracts from B16F10 cells, using SNRP70 Antibody. The lane on the left was treated with blocking peptide.)
Immunohistochemistry (IHC)-Paraffin
(MBS9612309 at 1/100 staining Rat brain tissue by IHC-P. The sample was formaldehyde fixed and a heat mediated antigen retrieval step in citrate buffer was performed. The sample was then blocked and incubated with the primary antibody at 4°C overnight. An HRP conjugated anti-Rabbit antibody was used as the secondary antibody.)
Immunofluorescence (IF)/Immunocytochemistry (ICC)
(MBS9612309 staining A549 cells by IF/ICC. The samples were fixed with PFA and permeabilized in 0.1% Triton X-100,then blocked in 10% serum for 45 minutes at 25°C. Samples were then incubated with primary Ab(MBS9612309) and mouse anti-beta tubulin Ab(T0023) for 1 hour at 37°C. An AlexaFluor594 conjugated goat anti-rabbit IgG(H+L) Ab(Red) and an AlexaFluor488 conjugated goat anti-mouse IgG(H+L) Ab(Green) were used as the secondary antibody. The nuclear counter stain is DAPI (blue).)
Post Translational Modifications: The N-terminus is blocked.Extensively phosphorylated on serine residues in the C-terminal region.
Subcellular Location: Nucleus speckle. Nucleus>Nucleoplasm. Note: Colocalizes with SCNM1 and LUC7L2 in nuclear speckles.
Subunit Structure: Component of the U1 snRNP (PubMed:19325628, PubMed:21113136, PubMed:25555158). The U1 snRNP is composed of the U1 snRNA and the 7 core Sm proteins SNRPB, SNRPD1, SNRPD2, SNRPD3, SNRPE, SNRPF and SNRPG that assemble in a heptameric protein ring on the Sm site of the small nuclear RNA to form the core snRNP, and at least three U1 snRNP-specific proteins SNRNP70/U1-70K, SNRPA/U1-A and SNRPC/U1-C (PubMed:19325628, PubMed:21113136, PubMed:25555158). Interacts with SCNM1 (By similarity). Found in a pre-mRNA splicing complex with SFRS4, SFRS5, SNRNP70, SNRPA1, SRRM1 and SRRM2 (PubMed:9531537). Found in a pre-mRNA exonic splicing enhancer (ESE) complex with SNRNP70, SNRPA1, SRRM1 and TRA2B/SFRS10 (PubMed:10339552). Interacts with dephosphorylated SFRS13A and SFPQ (PubMed:11514619, PubMed:14765198). Interacts with NUDT21/CPSF5, CPSF6, SCAF11, and ZRANB2 (PubMed:14561889, PubMed:11448987, PubMed:9447963). Interacts with GEMIN5 (PubMed:25911097). Interacts with FUS.
Similarity: The RRM domain mediates interaction with U1 RNA.

Uniprot Description
snRNP 70: mediates the splicing of pre-mRNA by binding to the loop I region of U1-snRNA. Participates in pre-mRNA splicing and exonic splicing complexes. A major ribonucleoprotein antigen recognized by the sera from patients with autoimmune diseases, such as systemic lupus erythematosus. Four alternatively spliced human isoforms have been described. The truncated isoforms (2-4) cannot bind U1-snRNA.

The SNRNP70 snrnp70 (Catalog #AAA9612309) is an Antibody produced from Rabbit and is intended for research purposes only. The product is available for immediate purchase. The SNRP70 Antibody reacts with Human, Mouse, Rat Predicted Reactivity: Zebrafish(86%), Bovine(100%), Sheep(100%), Rabbit(100%), Xenopus(100%) and may cross-react with other species as described in the data sheet. AAA Biotech's SNRP70 can be used in a range of immunoassay formats including, but not limited to, ELISA (EIA). WB: 1:500-1:2000 IF: 1:100-1:500 ICC: 1:100-1:500 IHC: 1:50-1:200 ELISA(peptide): 1:20,000-1:40,000. Researchers should empirically determine the suitability of the SNRNP70 snrnp70 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "SNRP70, Polyclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.Precautions
