Western Blot (WB)
(Western blot analysis of extracts from 3T3, using ZW10 Antibody. The lane on the left was treated with blocking peptide.)
Immunofluorescence (IF)/Immunocytochemistry (ICC)
(MBS9612153 staining Hela cells by IF/ICC. The samples were fixed with PFA and permeabilized in 0.1% Triton X-100,then blocked in 10% serum for 45 minutes at 25°C. Samples were then incubated with primary Ab(MBS9612153 1:200) and mouse anti-beta tubulin Ab(T0023 1:200) for 1 hour at 37°C. An AlexaFluor594 conjugated goat anti-rabbit IgG(H+L) Ab(Red) and an AlexaFluor488 conjugated goat anti-mouse IgG(H+L) Ab(Green) were used as the secondary antibody. The nuclear counter stain is DAPI(blue).)
Subcellular Location: Cytoplasm. Endoplasmic reticulum membrane>Peripheral membrane protein. Chromosome>Centromere>Kinetochore. Cytoplasm>Cytoskeleton>Spindle. Note: Dynamic pattern of localization during the cell cycle. In most cells at interphase, present diffusely in the cytoplasm (PubMed:15029241). In prometaphase, associated with the kinetochore. At metaphase, detected both at the kinetochores and, most prominently, at the spindle, particularly at the spindle poles. In very early anaphase, detected on segregating kinetochores. In late anaphase and telophase, accumulates at the spindle midzone (PubMed:11590237).
Tissue Specificity: Widely expressed.
Subunit Structure: Interacts with NBAS and KNTC1/ROD; the interactions are mutually exclusive and indicative for its association in two different vesicle tethering complexes (PubMed:11590237, PubMed:15824131, PubMed:20462495). Component of the RZZ complex composed of KNTC1/ROD, ZW10 and ZWILCH (PubMed:12686595, PubMed:20462495). Component of the NRZ complex composed of NBAS, ZW10 and RINT1/TIP20L; NRZ associates with SNAREs STX18, USE1L, BNIP1/SEC20L and SEC22B (the assembly has been described as syntaxin 18 complex). Interacts directly with RINT1/TIP20L bound to BNIP1/SEC20L (PubMed:15029241, PubMed:15272311, PubMed:20462495, PubMed:19369418). Interacts with C19orf25 and ZWINT (PubMed:15485811, PubMed:15824131., PubMed:16732327).
Similarity: Belongs to the ZW10 family.
NCBI and Uniprot Product Information
Predicted Molecular Weight: (Calculated)89kDa.
NCBI Description
This gene encodes a protein that is one of many involved in mechanisms to ensure proper chromosome segregation during cell division. This protein is an essential component of the mitotic checkpoint, which prevents cells from prematurely exiting mitosis. [provided by RefSeq, Aug 2011]
Uniprot Description
ZW10: Essential component of the mitotic checkpoint, which prevents cells from prematurely exiting mitosis. Required for the assembly of the dynein-dynactin and MAD1-MAD2 complexes onto kinetochores. Involved in regulation of membrane traffic between the Golgi and the endoplasmic reticulum. Belongs to the ZW10 family.

Product Notes
The ZW10 zw10 (Catalog #AAA9612153) is an Antibody produced from Rabbit and is intended for research purposes only. The product is available for immediate purchase. The ZW10 Antibody reacts with Human, Mouse, Rat Predicted Reactivity: Pig(100%), Bovine(100%), Horse(100%), Sheep(100%), Rabbit(100%), Dog(100%), Chicken(86%) and may cross-react with other species as described in the data sheet. AAA Biotech's ZW10 can be used in a range of immunoassay formats including, but not limited to, ELISA (EIA). WB: 1:500-1:2000 IF: 1:100-1:500 ICC: 1:100-1:500 ELISA(peptide): 1:20,000-1:40,000. Researchers should empirically determine the suitability of the ZW10 zw10 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "ZW10, Polyclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.Precautions
