Principle of the Assay: The LumiAb Human MIG Chemiluminescent ELISA Kit contains the components necessary for quantitative determination of natural or recombinant Human MIG concentrations within any experimental sample including cell lysates, serum and plasma. This particular immunoassay utilizes the quantitative technique of a "Sandwich" Enzyme-Linked Immunosorbent Assay (ELISA) where the target protein (antigen) is bound in a "sandwich" format by the primary capture antibodies coated to each well-bottom and the secondary detection antibodies added subsequently by the investigator. The capture antibodies coated to the bottom of each well are specific for a particular epitope on Human MIG while the user-added detection antibodies bind to epitopes on the captured target protein. Amid each step of the procedure, a series of wash steps must be performed to ensure the elimination of non-specific binding between proteins to other proteins or to the solid phase. After incubation and "sandwiching" of the target antigen, a peroxidase enzyme is conjugated to the constant heavy chain of the secondary antibody (either covalently or via Avidin/Streptavidin-Biotin interactions), allowing for a sensitive luminescent reaction to ensue upon substrate addition. When the Peroxide Enhancer solution is added, the reaction catalyzed by peroxidase yields light that is representative of the antigen concentration. After a brief incubation, the microplate can be read with a luminometer, allowing for generation of a standard curve and subsequent determination of protein concentration!!Background/Introduction: Human MIG or Monokine Induced by IFN-gamma, also known as C-X-C Motif Chemokine 9, is a 125 amino acid chemokine protein that affects the growth, movement or activation state of cells that participate in immune and inflammatory response. MIG is expressed from the CXCL9 gene located at locus 4q21 on chromosome 4. Via its binding with CXCR3, MIG is chemotactic for activated T-cells and is thought to be involved in T-cell trafficking.

NCBI Description

This locus represents a small inducible cytokine. The encoded protein, also known as macrophage inflammatory protein 1 alpha, plays a role in inflammatory responses through binding to the receptors CCR1, CCR4 and CCR5. Polymorphisms at this locus may be associated with both resistance and susceptibility to infection by human immunodeficiency virus type 1.[provided by RefSeq, Sep 2010]

Uniprot Description

CCL3: Monokine with inflammatory and chemokinetic properties. Binds to CCR1, CCR4 and CCR5. One of the major HIV-suppressive factors produced by CD8+ T-cells. Recombinant MIP-1-alpha induces a dose-dependent inhibition of different strains of HIV-1, HIV-2, and simian immunodeficiency virus (SIV). Belongs to the intercrine beta (chemokine CC) family.

Product Notes

The Human MIP-1alpha ccl3 (Catalog #AAA9510806) is a CLIA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA9510806 CLIA Kit recognizes Human MIP-1alpha. It is sometimes possible for the material contained within the vial of "MIP-1alpha, CLIA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
