Applicable Applications for FKBP1A sirna
RNA Interference (RNAi)
Quality Control
Oligonucleotide synthesis is monitored base by base through trityl analysis to ensure appropriate coupling efficiency. The oligo is subsequently purified by affinity-solid phase extraction. The annealed RNA duplex is further analyzed by mass spectrometry to verify the exact composition of the duplex. Each lot is compared to the previous lot by mass spectrometry to ensure maximum lot-to-lot consistency.
Directions for Use
We recommends transfection with 100 nM siRNA 48 to 72 hours prior to cell lysis. Before resuspending, briefly centrifuge the tube to ensure the lyophilized siRNA is at the bottom of the tube. Resuspend the siRNA oligos to an appropriate concentration with DEPC water. For each vial, suitable for 250 transfections in 24 well plate (20 pmol for each well).
Components
We offer pre-designed sets of 3 different target-specific siRNA oligo duplexes of mouse FKBP1A gene. Each vial contains 5 nmol of lyophilized siRNA. The duplexes can be transfected individually or pooled together to achieve knockdown of the target gene, which is most commonly assessed by qPCR or western blot. Our siRNA oligos are also chemically modified (2'-OMe) at no extra charge for increased stability and enhanced knockdown in vitro and in vivo.
Preparation and Storage
Store at -20 degree C for one year.

NCBI Description

This gene is a member of the immunophilin family. The encoded protein is a cis-trans prolyl isomerase that binds the immunosuppressants FK506 and rapamycin, and is associated with immunoregulation, protein folding, receptor signaling, protein trafficking and T-cell activation. It may modulate the calcium release activity of the ryanodine receptor Ryr1. It also interacts with the type I TGF-beta receptor. Disruption of this gene in mouse causes severe ventricular defects. Pseudogenes of this gene have been defined on chromosomes 4, 10, 14, and 16. Alternative splicing results in multiple transcript variants. [provided by RefSeq, Sep 2014]

Uniprot Description

FKBP12: an immunophilin and peptidyl-prolyl cis-trans isomerase that binds to the immunosuppressant drugs FK506 (a.k.a. tacrolimus or Fujimycin) and rapamycin (a.k.a. Sirolimus). The FKBP12/rapamycin complex binds NFAT transcription factors and inhibits the induction of T cell genes including IL-2, IL-3, IL-4, TNF-alpha and GM-CSF. FK506 is used in treating patients after organ transplant and those suffering from autoimmune disorders. The immunosuppressant activity of FKBP12 is not apparently related to its prolyl isomerase activity. The FKBP12/rapamycin complex inhibits the mammalian target of rapamycin (mTOR) pathway by directly binding the mTOR Complex1 (mTORC1). mTORC1 contains Raptor, mLST8, and PRAS40, and interacts with DEPTOR, which inhibits its activity. mTOR, a S/T protein kinase, is a central regulator of cellular growth and metabolism. Its inhibition enhances the dependence on aerobic glycolysis in leukemic cells.

Product Notes

The FKBP1A fkbp1a (Catalog #AAA828748) is a siRNA produced from Synthetic and is intended for research purposes only. The product is available for immediate purchase. The FKBP1A siRNA (Mouse) reacts with Mouse and may cross-react with other species as described in the data sheet. AAA Biotech's FKBP1A can be used in a range of immunoassay formats including, but not limited to, RNA Interference (RNAi). Researchers should empirically determine the suitability of the FKBP1A fkbp1a for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "FKBP1A, siRNA" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
