Immunohistochemistry (IHC)

(Immunohistochemical analysis of TRAP100 staining in human brain formalin fixed paraffin embedded tissue section. The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH 6.0). The section was then incubated with the antibody at room temperature and detected using an HRP conjugated compact polymer system. DAB was used as the chromogen. The section was then counterstained with haematoxylin and mounted with DPX.)

Immunofluorescence (IF)

(Immunofluorescent analysis of TRAP100 staining in HepG2 cells. Formalin-fixed cells were permeabilized with 0.1% Triton X-100 in TBS for 5-10 minutes and blocked with 3% BSA-PBS for 30 minutes at room temperature. Cells were probed with the primary antibody in 3% BSA-PBS and incubated overnight at 4 °C in a humidified chamber. Cells were washed with PBST and incubated with a DyLight 594-conjugated secondary antibody (red) in PBS at room temperature in the dark.)

NCBI Description

This gene encodes a component of the mediator complex (also known as TRAP, SMCC, DRIP, or ARC), a transcriptional coactivator complex thought to be required for the expression of almost all genes. The mediator complex is recruited by transcriptional activators or nuclear receptors to induce gene expression, possibly by interacting with RNA polymerase II and promoting the formation of a transcriptional pre-initiation complex. Multiple transcript variants encoding different isoforms have been found for this gene. [provided by RefSeq, Jul 2008]

Uniprot Description

Trap100: a transcriptional coactivator protein of the thyroid hormone receptor-associated protein (TRAP) family. Forms a complex with the thyroid hormone receptor (TR) and markedly enhance TR-mediated transcription in vitro. Interacts and co-precipitates with Trap220 but does not directly bind nuclear hormone receptors.

Protein type: Transcription, coactivator/corepressor; Nuclear receptor co-regulator

Chromosomal Location of Human Ortholog: 17q21.1

Cellular Component: nucleoplasm; nucleus; Srb-mediator complex; ubiquitin ligase complex

Molecular Function: histone acetyltransferase activity; ligand-dependent nuclear receptor transcription coactivator activity; protein binding; receptor activity; thyroid hormone receptor binding; transcription cofactor activity; vitamin D receptor binding

Biological Process: androgen receptor signaling pathway; gene expression; histone acetylation; positive regulation of transcription, DNA-dependent; protein heterooligomerization; protein ubiquitination; regulation of transcription from RNA polymerase II promoter; stem cell maintenance; steroid hormone receptor signaling pathway; transcription initiation from RNA polymerase II promoter

Product Notes

The MED24 med24 (Catalog #AAA8242661) is an Antibody produced from Rabbit and is intended for research purposes only. The product is available for immediate purchase. The Anti-TRAP100 Antibody reacts with Human, Rat, Bovine, Chicken, Pig and may cross-react with other species as described in the data sheet. AAA Biotech's TRAP100 can be used in a range of immunoassay formats including, but not limited to, Western Blot (WB), Immunohistochemistry (IHC), Immunofluorescence (IF), Immunocytochemistry (ICC). Western Blot: (1/500 - 1/1000); Immunohistochemistry: (1/100 - 1/200); Immunofluorescence/Immunocytochemistry: (1/100 - 1/500). Researchers should empirically determine the suitability of the MED24 med24 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "TRAP100, Polyclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
