This gene encodes a protein localized to the tight junctions and adherens junctions in vertebrate epithelial cells. The encoded protein regulates the activity of Rho family GTPases during junction assembly and at confluence. At the adherens junctions, the encoded protein is part of a protein complex that links E-cadherin to the microtubule cytoskeleton. Alternative splicing of this gene results in multiple transcript variants. [provided by RefSeq, Jan 2015]

CGNL1: May be involved in anchoring the apical junctional complex, especially tight junctions, to actin-based cytoskeletons. Defects in CGNL1 are a cause of aromatase excess syndrome (AEXS). A disorder characterized by an estrogen excess due to an increased aromatase activity. A chromosomal aberration inv(15)(q21.2;q21.3) has been found in patients with aromatase excess syndrome. The inversion moves the promoter of the CGNL1 gene into a 5-prime position in relation to the aromatase coding region. Belongs to the cingulin family. 2 isoforms of the human protein are produced by alternative splicing.
