Applicable Applications for POLR2J sirna
RNA Interference (RNAi)
Quality Control
Oligonucleotide synthesis is monitored base by base through trityl analysis to ensure appropriate coupling efficiency. The oligo is subsequently purified by affinity-solid phase extraction. The annealed RNA duplex is further analyzed by mass spectrometry to verify the exact composition of the duplex. Each lot is compared to the previous lot by mass spectrometry to ensure maximum lot-to-lot consistency.
Directions for Use
We recommends transfection with 100 nM siRNA 48 to 72 hours prior to cell lysis. Before resuspending, briefly centrifuge the tube to ensure the lyophilized siRNA is at the bottom of the tube. Resuspend the siRNA oligos to an appropriate concentration with DEPC water. For each vial, suitable for 250 transfections in 24 well plate (20 pmol for each well).
Components
We offer pre-designed sets of 3 different target-specific siRNA oligo duplexes of human POLR2J gene. Each vial contains 5 nmol of lyophilized siRNA. The duplexes can be transfected individually or pooled together to achieve knockdown of the target gene, which is most commonly assessed by qPCR or western blot. Our siRNA oligos are also chemically modified (2'-OMe) at no extra charge for increased stability and enhanced knockdown in vitro and in vivo.
Preparation and Storage
Shipped at 4 degree C. Store at -20 degree C for one year.

NCBI Description

This gene encodes a subunit of RNA polymerase II, the polymerase responsible for synthesizing messenger RNA in eukaryotes. The product of this gene exists as a heterodimer with another polymerase subunit; together they form a core subassembly unit of the polymerase. Two similar genes are located nearby on chromosome 7q22.1 and a pseudogene is found on chromosome 7p13. [provided by RefSeq, Jul 2008]

Uniprot Description

POLR2J: DNA-dependent RNA polymerase catalyzes the transcription of DNA into RNA using the four ribonucleoside triphosphates as substrates. Component of RNA polymerase II which synthesizes mRNA precursors and many functional non-coding RNAs. Pol II is the central component of the basal RNA polymerase II transcription machinery. It is composed of mobile elements that move relative to each other. RPB11 is part of the core element with the central large cleft. Belongs to the archaeal RpoL/eukaryotic RPB11/RPC19 RNA polymerase subunit family.

Protein type: RNA splicing; DNA-binding; RNA processing; Transferase

Chromosomal Location of Human Ortholog: 7q22.1

Cellular Component: nucleoplasm; DNA-directed RNA polymerase II, core complex; nucleus

Molecular Function: protein dimerization activity; protein binding; DNA binding; DNA-directed RNA polymerase activity; LRR domain binding

Biological Process: transcription from RNA polymerase II promoter; nuclear mRNA splicing, via spliceosome; transcription initiation from RNA polymerase II promoter; mRNA capping; viral reproduction; somatic stem cell maintenance; nucleotide-excision repair; positive regulation of viral transcription; RNA splicing; transcription-coupled nucleotide-excision repair; RNA elongation from RNA polymerase II promoter; gene expression; DNA repair

Product Notes

The POLR2J polr2j (Catalog #AAA8211941) is a siRNA produced from Synthetic and is intended for research purposes only. The product is available for immediate purchase. The POLR2J siRNA (Human) reacts with Human and may cross-react with other species as described in the data sheet. AAA Biotech's POLR2J can be used in a range of immunoassay formats including, but not limited to, RNA Interference (RNAi). Researchers should empirically determine the suitability of the POLR2J polr2j for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "POLR2J, siRNA" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
