Principle of the Assay: This kit was based on sandwich enzyme-linked immune-sorbent assay technology. Anti- AMH antibody was pre-coated onto 96-well plates. And the biotin conjugated anti- AMH antibody was used as detection antibodies. The standards, test samples and biotin conjugated detection antibody were added to the wells subsequently, and washed with wash buffer. HRPStreptavidin was added and unbound conjugates were washed away with wash buffer. TMB substrates were used to visualize HRP enzymatic reaction. TMB was catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. The density of yellow is proportional to the AMH amount of sample captured in plate. Read the O.D. absorbance at 450nm in a microplate reader, and then the concentration of AMH can be calculated.

Anti-Mullerian hormone is a member of the transforming growth factor-beta gene family which mediates male sexual differentiation. Anti-Mullerian hormone causes the regression of Mullerian ducts which would otherwise differentiate into the uterus and fallopian tubes. AMH: This glycoprotein, produced by the Sertoli cells of the testis, causes regression of the Muellerian duct. It is also able to inhibit the growth of tumors derived from tissues of Muellerian duct origin. Defects in AMH are the cause of persistent Muellerian duct syndrome type 1 (PMDS1). PMDS1 is a form of male pseudohermaphroditism characterized by a failure of Muellerian duct regression in otherwise normal males. Belongs to the TGF-beta family.
