Principle of the Assay||This kit was based on sandwich enzyme-linked immune-sorbent assay technology. Anti-Human TNFSF14 antibody was pre-coated onto 96-well plates. And the biotin conjugated anti-Human TNFSF14 antibody was used as detection antibodies. The standards, test samples and biotin conjugated detection antibody were added to the wells subsequently, and wash with wash buffer. HRP-Streptavidin was added and unbound conjugates were washed away with wash buffer. TMB substrates were used to visualize HRP enzymatic reaction. TMB was catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. The density of yellow is proportional to the Human TNFSF14 amount of sample captured in plate. Read the O.D. absorbance at 450nm in a microplate reader, and then the concentration of Human TNFSF14 can be calculated.

NCBI Description

The protein encoded by this gene is a member of the tumor necrosis factor (TNF) ligand family. This protein is a ligand for TNFRSF14, which is a member of the tumor necrosis factor receptor superfamily, and which is also known as a herpesvirus entry mediator (HVEM). This protein may function as a costimulatory factor for the activation of lymphoid cells and as a deterrent to infection by herpesvirus. This protein has been shown to stimulate the proliferation of T cells, and trigger apoptosis of various tumor cells. This protein is also reported to prevent tumor necrosis factor alpha mediated apoptosis in primary hepatocyte. Two alternatively spliced transcript variant encoding distinct isoforms have been reported. [provided by RefSeq, Jul 2008]

Uniprot Description

TNFSF14: Cytokine that binds to TNFRSF3/LTBR. Binding to the decoy receptor TNFRSF6B modulates its effects. Activates NFKB, stimulates the proliferation of T-cells, and inhibits growth of the adenocarcinoma HT-29. Acts as a receptor for Herpes simplex virus. Belongs to the tumor necrosis factor family. 2 isoforms of the human protein are produced by alternative splicing.

Protein type: Membrane protein, integral; Cytokine; Inhibitor

Chromosomal Location of Human Ortholog: 19p13.3

Cellular Component: cytoplasm; extracellular space; integral to membrane; plasma membrane

Molecular Function: caspase inhibitor activity; cytokine activity; protein binding; receptor binding; tumor necrosis factor receptor binding

Biological Process: apoptosis; immune response; negative regulation of caspase activity; positive regulation of myoblast differentiation; release of cytoplasmic sequestered NF-kappaB; signal transduction; T cell activation; T cell costimulation; T cell homeostasis; T cell proliferation; tumor necrosis factor-mediated signaling pathway

Product Notes

The Human TNFSF14/LIGHT tnfsf14 (Catalog #AAA760735) is an ELISA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA760735 ELISA Kit recognizes Human TNFSF14/LIGHT. It is sometimes possible for the material contained within the vial of "Tumor necrosis factor ligand superfamily member 14, ELISA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
