Sheep Ribonuclease T2 ELISA Kit
Principle of the Assay: RNASET2 ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-RNASET2 antibody and an RNASET2-HRP conjugate. The assay sample and buffer are incubated together with RNASET2-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the RNASET2 concentration since RNASET2 from samples and RNASET2-HRP conjugate compete for the anti-RNASET2 antibody binding site. Since the number of sites is limited, as more sites are occupied by RNASET2 from the sample, fewer sites are left to bind RNASET2-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The RNASET2 concentration in each sample is interpolated from this standard curve.

RNASET2: Has ribonuclease activity, with higher activity at acidic pH. May play a role in cellular RNA catabolism. Defects in RNASET2 are the cause of leukoencephalopathy cystic without megalencephaly (LCWM). An infantile- onset syndrome of cerebral leukoencephalopathy. Affected newborns develop microcephaly and neurologic abnormalities including psychomotor impairment, seizures and sensorineural hearing impairment. The brain shows multifocal white matter lesions, anterior temporal lobe subcortical cysts, pericystic abnormal myelination, ventriculomegaly and intracranial calcifications. Belongs to the RNase T2 family. 2 isoforms of the human protein are produced by alternative splicing.
