Principle of the assay: IL-6R ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-IL-6R antibody and an IL-6R-HRP conjugate. The assay sample and buffer are incubated together with IL-6R-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the IL-6R concentration since IL-6R from samples and IL-6R-HRP conjugate compete for the anti-IL-6R antibody binding site. Since the number of sites is limited, as more sites are occupied by IL-6R from the sample, fewer sites are left to bind IL-6R-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The IL-6R concentration in each sample is interpolated from this standard curve.

NCBI Description

This gene encodes a subunit of the interleukin 6 (IL6) receptor complex. Interleukin 6 is a potent pleiotropic cytokine that regulates cell growth and differentiation and plays an important role in the immune response. The IL6 receptor is a protein complex consisting of this protein and interleukin 6 signal transducer (IL6ST/GP130/IL6-beta), a receptor subunit also shared by many other cytokines. Dysregulated production of IL6 and this receptor are implicated in the pathogenesis of many diseases, such as multiple myeloma, autoimmune diseases and prostate cancer. Alternatively spliced transcript variants encoding distinct isoforms have been reported. A pseudogene of this gene is found on chromosome 9.[provided by RefSeq, May 2011]

Type 3 subfamily. 2 isoforms of the human protein are produced by alternative splicing.
