Principle of the assay: C8b ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-C8b antibody and an C8b-HRP conjugate. The assay sample and buffer are incubated together with C8b-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the C8b concentration since C8b from samples and C8b-HRP conjugate compete for the anti-C8b antibody binding site. Since the number of sites is limited, as more sites are occupied by C8b from the sample, fewer sites are left to bind C8b-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The C8b concentration in each sample is interpolated from this standard curve.

This gene encodes one of the three subunits of the complement component 8 (C8) protein. C8 is composed of equimolar amounts of alpha, beta and gamma subunits, which are encoded by three separate genes. C8 is one component of the membrane attack complex, which mediates cell lysis, and it initiates membrane penetration of the complex. This protein mediates the interaction of C8 with the C5b-7 membrane attack complex precursor. In humans deficiency of this protein is associated with increased risk of meningococcal infections.
